Bottleneck removal of paclitaxel biosynthetic pathway by overexpression of DBTNBT gene under methyl-?-cyclodextrin and coronatine elicitation in Taxus baccata L.

Paclitaxel is a highly functionalized diterpenoid that broadly used for the treatment of several cancer types. This valuable specialized metabolite naturally exists in inner bark Taxus species low amounts. The limited-scale production paclitaxel cell cultures has necessitated use elicitors. Recently, methyl-?-cyclodextrin (CD) and coronatine (COR) have been considered to be effective elicitors producing plant metabolites. Given limited due rate limiting enzymes' function, bottleneck removal conducive more significant amounts paclitaxel. In present study, full length DBTNBT coding sequence (CDS), as one pathway bottlenecks, was integrated downstream CaMV 35S promoter (pCAMBIA1304-DBTNBT) transiently expressed baccata leaves via Agrobacterium tumefaciens vacuum infiltration method. expression level involved genes were evaluated through different treatments. transient overexpression gene, associated with dual elicitation, resulted 7.4-fold compared no-inoculation/no-elicitation control.
